RNA Synthesis
The RNA Medicine Core offers both reporter mRNAs and customizable RNA synthesis services. Our reporter gene mRNAs include EGFP, EYFP, mCherry, FFLuc, Renilla Luc, and Nano Luc. We provide 100 up to 15 mg of Cap-1 mRNAs and mRNAs with 5-methoxyuridine (5moU) and N1-methylpseudouridine (N1MePsU) modification. These mRNAs are capped and polyadenylated, making them ready for transfection and expression in vitro (e.g. < 100 ug) or in vivo (e.g. > 300 ug), providing flexibility and scalability for your diverse research needs both in vitro and in vivo.

EGFP mRNA
The EGFP mRNA produces a green fluorescent protein (26.94kDa) with an excitation peak at 488 nm and an emission peak at 509 nm.

EYFP mRNA
The EYFP mRNA produces yellow-green fluorescent protein(27.07kDa) with an excitation peak at 516 nm and an emission peak at 527 nm.

mCherry mRNA
The mCherry mRNA produces a red fluorescent protein (26.72kDa) with an excitation peak at 587 nm and an emission peak at 610 nm.

FFLuc mRNA
Firefly Luciferase (FFluc, 60.64kDa) uses D-Luciferin as substrate and emits light in the yellow-green spectrum, peaks ~560 nm.

Renilla Luc mRNA
Renilla luciferase (Rluc, 35.99kDa) uses coelenterazine as its substrate and emits light in the blue-green spectrum, peaks ~480 nm.

NanoLuc mRNA
NanoLuc luciferase (Nluc, 19.09kDa) uses furimazine as substrate, which produces high-intensity, glow-type luminescence. NanoLuc generates brighter signal compared to Firefly and Renilla luciferases.

OVA mRNA
Ovalbumin (OVA, 45kDa) is a protein often used as a model antigen in immunological research. It helps scientists study immune responses and develop vaccines.

PD-L1 mRNA
Programmed death-ligand 1 (PD-L1), a type I transmembrane protein (~ 40 kDa), plays a key role in immune regulation by binding to the PD-1 receptor on T cells to inhibit immune responses.

IL2RB mRNA
Interleukin-2 receptor subunit beta (IL2RB), a ~40 kDa type I membrane glycoprotein, mediates IL-2/IL-15 signaling and has been explored as a vaccine target to enhance T-cell activation and immune responses.

Bioluminescence Assays
For detecting Firefly Luciferase (FFLuc), Renilla Luciferase (Rluc), and NanoLuc Luciferase (Nluc) in mRNA bioluminescence assays, we recommend using these kits:
1. Firefly Luciferase (FFLuc):
ONE-Glo® Luciferase Assay System: This kit allows highly sensitive, robust detection of firefly luciferase reporter gene expression in mammalian cells. It's widely used for its sensitivity and ease of use.
2. Renilla Luciferase (Rluc):
